Etymology

Literally, “dick in(to) the asshole”. The noun sense (“in”) is a metaphor for pain and discomfort, painting the obnoxious person as being a pain in the ass; the interjection sense (“into [one's]”) uses expressive anal rape, wished upon the target of insult, as a means of offense. (Compare English get fucked, which similarly wishes for rape upon the insulted.) See also vai tomar no cu (“fuck off! up yours!”, literally “go and take it up the asshole”).
